Overview

Poly [ADP-ribose] polymerase 15 (PARP15) is a member of the PARP family of proteins, which modify target proteins by ADP-ribosylation, a reversible post-translational modification using NAD+ as a substrate[1][2][4]. PARP15 is characterized by the presence of a macro domain and catalyzes mono-ADP-ribosylation (not poly)[2][4]. Its physiological functions are poorly understood, but PARP15 is grouped within the “macro-PARP” subfamily (alongside PARP9 and PARP14), suggesting possible regulatory roles in transcription and immune signaling, as well as associations with cancer development or prognosis[2][4]. Structural and inhibitor studies exist, but no clinically approved drugs are specific for PARP15, and its role as a pharmacological target remains mostly exploratory[3][2][4].
